The Trail does not pass through the grounds of Derrinal Station. To do so would have required two highway crossings and access to what is now private property. It is routed a short distance along Derrinal Station Lane, through a lightly timbered area to the Derrinal Rest Area and continuing over what was McIvor Road, now Old Bendigo Road, to join the rail line at a steel bridge closer to Heathcote. A replica station sign has been placed near to the Trail at the rest area with facilities. The original Derrinal Station passenger platform can be seen across the highway from beneath the sign.
The Trail rejoins the rail alignment a little beyond the intersection of Old Bendigo Road and McIvor Road, at the Wallace Gully Bridge, opposite the current day entrance to Kilmuir Estate.
